dc.description.abstract | Thelarge-scaleminingandhigh-throughputgenotypingofnovelgene-basedallelicvariantsinnaturalmappingpopulationareessentialforassociationmappingtoidentifyfunctionallyrelevantmoleculartagsgoverningusefulagronomictraitsinchickpea.Thepresentstudyemploysanalternativetime-saving,non-laboriousandeconomicalpool-basedEcoTILLINGapproachcoupledwithagarosegeldetectionassaytodiscover1133novelSNPallelicvariantsfromdiversecodingandregulatorysequencecomponentsof1133transcriptionfactor(TF)genesbygenotypingin192diversedesiandkabulichickpeaaccessionsconstitutingaseedweightassociationpanel.IntegratingtheseSNPgenotypingdatawithseedweightfieldphenotypicinformationof192structuredassociationpanelidentifiedeightSNPallelesintheeightTFgenesregulatingseedweightofchickpea.TheassociatedindividualandcombinationofallSNPsexplained10–15and31%phenotypicvariationforseedweight,respectively.TheEcoTILLING-basedlarge-scalealleleminingandgenotypingstrategyimplementedforassociationmappingisfoundmucheffectiveforadiploidgenomecropspecieslikechickpeawithnarrowgeneticbaseandlowgeneticpolymorphism.Thisoptimizedapproachthuscanbedeployedforvariousgenomics-assistedbreedingapplicationswithoptimalexpenseofresourcesindomesticatedchickpea.Theseedweight-associatednaturalallelicvariantsandcandidateTFgenesdelineatedhavepotentialtoacceleratemarker-assistedgeneticimprovementofchickpea. | en_US |
